镍激活EB病毒的研究

Study on Activation of Ni on Epstein-Barr Virus

应用间接免疫荧光法和抗补体免疫荧光技术研究 ,证实在试管内 1～ 4 0mg/LNiSO4 ·6H2 O (Ni)显著增强携带Epstein -BarrVirus (EBV)基因组的三株类淋巴母细胞株的VCA、MA、EA、EBNA - 2抗原表达。此外还发现 1～ 2 0mg/LNi能明显促进EBV转化人脐血B淋巴细胞的作用 ,当多次加Ni时促进效应更强 ,前者转化促进率为 13 31%～ 4 6 6 1% ,后者达 2 2 70 %～ 57 0 4 %。以上结果表明 ,Ni具有激活EBV的作用。EBV与鼻咽癌 (NPC)密切相关 ,广东NPC高发区大米和饮水中Ni的含量偏高 ,NPC患者血和头发中Ni的水平明显高于健康人。研究结果提示 ,EBV与Ni相互作用可能是NPC发生、发展过程中的一个协同因素。

The activation of Ni on Epstein-Barr virus was studied by indirect immunofluerescence and anticomplement immunofluerescence staining .The results demonstrated that NiSO 4·6H 2O(Ni)(1～40 mg/L)can markedly enhance Epstein-Barr virus(EBV) VCA,MA,EA,EBNA-2 antigens expression in three EBV genome-positive lymphobiastaid cell lines and the transformation effect of B lymphocyte from human umbilical cord blood (CB) by EBV could be increased by 13 31%～46 61% in vitro by Ni(1～20 mg/L).The effect of Ni(1～20 mg/L)on CB transformation by EBV was signifiantly enhanced by 22 07%～57 04% when Ni was added at multiple times.These results indicated that Ni can activate EBV. EBV is associated with nasopharyngeal carcinoma(NPC).The content of Ni in rice and drinking water was significantly higher in high risk areas for NPC of Guangdong than that in low risk areas.The levels of Ni in blood and hair for NPC patients were markedly higher than those in healthy persons. The results suggest that the interaction of Ni and EBV may be a contributive confactor to the occurrence and development of NPC.
