摘要
目的研究脊髓损伤(SCI)后突触后密度蛋白93(PSD-93)的表达和分布。方法 54只SD大鼠建立SCI模型(A组),分别在SCI后8h、1、3、5、7、14d取材;另选9只大鼠为对照组(C组)。采用定量PCR和Western blot方法分别检测PSD-93mRNA和蛋白表达,免疫组织化学染色法检测PSD-93在正常和损伤脊髓中的分布。结果与C组相比,SCI后PSD-93mRNA表达下降,3d时达最低(P<0.01)。而SCI后1d时PSD-93蛋白表达高于C组(P<0.01),但随后逐渐下降,5d时降到最低(P<0.05)。PSD-93在正常脊髓中高表达在前角运动神经元和后角的浅表板层;SCI后1d时PSD-93在前角的阳性染色面积较C组增加(P<0.01)。结论 PSD-93可能参与脊髓的继发性损伤。
Objective To explore the expression and distribution of postsynaptic density-93 (PSD-93) after spinal cord injury(SCI) in rats. Methods SCI model was established in 54 SD rats (group A) and the spinal cord tissues were extracted at 8 h and on the 1st , 3rd , 5th , 7th and 14th day after SCI. Another 9 rats were taken as normal controls (group C). The mRNA and protein expressions of PSD-93 were detected by quantitative-PCR and Western blot, respectively. The distribution of PSD-93 in normal and injured spinal cord was measured by immunohistoehemistry. Results Compared with group C, the mRNA expression of PSD-93 was decreased after SCI with a minimum decrease on the 3rd day in group A(P〈0. 01), while the protein expression of PSD-93 was peaked on the 1st day and declined with a minimum decrease on the 5th day after SCI in group A (P〈0, 05). PSD-93 was highly expressed in the motoneurons of ventral horn and superficial lamina of dorsal horn of normal spinal cord. The positively stained areas of PSD-93 in the ventral horn on the 1st day after SCI in group A was larger than that in group C(P〈0. 01). Conclusion PSD-93 may participate in the secondary SCI.
