摘要 目的探讨c-Myc基因的表达与人非小细胞肺癌细胞增殖的关系。方法体外培养H1299细胞,分为空白对照(A组)、细胞对照(B组)、脂质体(C组)、小干扰RNA对照(D组)和c-Myc小干扰RNA组(E组)。采用实时荧光定量PCR检测c-Myc mRNA表达,细胞计数试剂盒(CCK-8)法检测细胞增殖情况。结果 c-Myc siRNA瞬时转染后48h呈浓度依赖性地抑制H1299细胞中c-Myc RNA的表达。与B、C、D组比较,E组细胞增殖率降低(P<0.05)。结论低表达c-Myc基因能抑制非小细胞肺癌H1299细胞的增殖。 Objective To investigate the relationship between c-Myc expression and proliferation of non-small cell lung cancer(NSCLC) cells. Methods The H1299 cells were cultured in vitro and divided into five groups of A(blank control), B(H1299 cells alone), C (treated with Lipofectamine 2000),D(transfected with control siRNA) and E(transfected with c-Myc siRNA). The mRNA expression of c-Myc was detected by RT-PCR and the proliferation of H1299 cells was determined by CCK-8. Results After H1299 cells were transiently transfected with c-Myc siRNA for 48 hours,the mRNA expression of c-Myc was inhibited in a dose-dependent manner. Compared with groups of B,C and D, the proliferation rate of H1299 cells was decreased in group E(P〈0. 05). Conclusion Downreulation of c-Mvc exor'ession can inhibit the proliferation of NSCLC H1299 cells.
