烟酰胺与全反式维A酸相互作用对小鼠表皮结构的影响 被引量:1

摘要 目的研究烟酰胺与全反式维A酸相互作用对小鼠皮肤表皮结构的影响,明确烟酰胺是否具有减弱全反式维A酸所引起的小鼠表皮结构改变的作用。方法将小鼠背部皮肤分为4个区域,分别外用0.1%全反式维A酸(ATRA)乳剂、4%烟酰胺(Nic)乳剂、0.1%全反式维A酸联合4%烟酰胺及乳膏基质5d,通过HE染色及透射电镜观察小鼠表皮组织结构及超微结构的变化。结果全反式维A酸组小鼠表皮组织结构异常,出现角化不全、细胞间水肿、真皮浅中层炎症细胞浸润;超微结构中角质层破坏,细胞间隙增大;联合烟酰胺后小鼠皮肤组织结构仅有基底细胞间轻度水肿和少量真皮炎症细胞浸润;超微结构中表皮结构基本正常,细胞间隙也较维A酸组小。结论全反式维A酸破坏小鼠表皮结构,烟酰胺可减弱其对表皮结构的破坏作用。 Objective To investigate the the effects of niacinamide (Nic) and alltransretinoic acid (ATRA) on epi dermis structure in mice, so that we can well define whether Nic could be used to reversed the side effect of ATRA on epidermis structure. Methods Firstly,0.1% ATRA,4% Nic ,0.1% ATRA with 4% Nic were ap plied separately on mice back which was divided into four sections, then the epidermis structures were ob served by HE staining and electromicroscope. Results We found abnormal histopathological changes in the group treated by ATRA, including parakeratosis, intercellular edema and dermal inflammatory cell infiltra tion. The ultrastructure reflected the damage of stratum corneum that intercellular space increased, but the group of combined with niacinamide alleviate a lot than ATRA treated alone, on the aspects such as intercel lular edema,dermal inflammatory cell infiltration and intercellular space. Conclusion ATRA destroyed the epidermis structure and niacinamide reversed the side effects of ATRA on epidermis structure.
