摘要
目的探讨白藜芦醇对缺血再灌注大鼠海马谷氨酸的影响。方法建立大鼠局灶性脑缺血再灌注模型,将30只雄性SD大鼠随机分成假手术组、模型组和白藜芦醇组,每组10只。模型组、白藜芦醇组在再灌注前5 min分别静脉注射0.04%丙二醇溶液、白藜芦醇溶液。反相高效液相层析(RP-HPLC)检测缺血再灌注大鼠海马组织匀浆谷氨酸含量,免疫组化SP法测定谷氨酸转运蛋白EAAC-1表达。结果白藜芦醇组大鼠神经功能缺损改善明显高于模型组,两者具有显著性差异(P<0.05);白藜芦醇组谷氨酸浓度较模型组明显降低,两者有极显著性差异(P<0.01);白藜芦醇组EAAC-1表达高于模型组,两者具有显著性差异(P<0.05)。结论白藜芦醇能够提高缺血再灌注模型大鼠神经元EAAC-1表达,加速细胞间隙谷氨酸清除,这可能是其减轻脑缺血再灌注损伤的重要作用机制之一。
Objective To evaluate the effect ofresveratrol on hippocampal glutamate level in ischemia reperfusion rats. Methods The ischemia reperfusion model of rats were established. Thirty male Sprague-Dawley rats were randomly divided into 3 groups: Sham-operate group (n = 10), model group (n = 10) and resveratrol group (n = 10). Model group and resveratrol group rats were given 0.04% propylene glycol solution and resveratrol solution respectively 5 min before reperfusion. The glutamate concentration in the ischemic brain tissue homogenate and the expression of glutamate transporter EAAC-1 were determined by reverse phase high-performance liquid chromatography (RP-HPLC) and immunohistochemistry (SP method) respectively. Results Compared with the model group, the improvement of neurological deficit was more significant with a significant difference (P 〈 0.05), the glutamate concentration was lower, with a statistically significant difference (P 〈 0.01), the expression of glutamate transporter EAAC-1 was higher, with a significant difference (P 〈 0.05) in resveratrol group. Conclusions Resveratrol can improve the expression of glutamate transporter EAAC-1 in the neuron and accelerate the elimination of glutamate in extracellular space, which might play an important role in alleviating ischemia reperfusion injury.
