17β-雌二醇对诱导绵羊输卵管上皮细胞β-防御素-1(SBD-1)基因表达的影响 被引量:1

Effects of 17β-estradiol on the Induced Sheep β-defensin-1 (SBD-1) Expression in the Ovine Oviduct Epithelial Cells

摘要 研究了17β-雌二醇(E2)对培养的绵羊输卵管上皮细胞内SBD-1基因表达的影响.将不同剂量的17β-雌二醇加入传2代的绵羊输卵管上皮细胞内,分为实验组(10-6 mol/L组、10-7 mol/L组、10-8 mol/L组、10-9 mol/L组和10-10 mol/L组)和相应的对照组,分别处理2,6,12,24,48h,然后通过实时荧光定量RT-PCR技术,检测17β-雌二醇对绵羊输卵管上皮细胞内SBD-1mRNA的表达变化.结果显示:17β-雌二醇对绵羊输卵管上皮细胞中SBD-1mRNA的表达存在着时间效应和剂量上的依赖关系;17β-雌二醇可以上调绵羊输卵管上皮细胞中SBD-1mRNA表达,上调表达的最佳时间为6h,最佳浓度为10-8 mol/L(P<0.01). To investigate the effect of 17β-estradiol (E2) on β-defensin-1 gene expression in the ovine oviduct epithelial cells,the 2th generation of the sheep oviduct epithelial cells were treated in different time (2,6,12,24,48 h) with various dose of 17β-estradiol (10.6 ,10.7 ,10-8 ,10-9 ,10 -10 mol/L),while a control group(without the 17β - estradiol,Control), then the level of SBD-1 gene expression in the ovine oviduct epithelial cells was detected by real-time RT-PCR. The results showed that 17β-estradiol regulated SBD-1 mRNA expression in a time and dose-dependent manner. It is said that 17β-estradiol significantly up-regu- lated SBD-1 mRNA expression, and the maximal stimulatory effect of 17β-estradiol on the expression of SBD-1 mRNA was observed at the concentration of 10-8 mol/L after treatment for 6 h(P〈0.01).
