摘要
目的研究自然杀伤细胞G2D受体在乙肝患者体内差异表达及其临床意义。方法选取2012年5月~2013年5月在本院就诊的30例慢性肝炎患者,21例重型肝炎患者及非乙肝对照18例,用流式细胞仪检测外周血G2D表达,采用免疫组化S-P法检测肝组织G2D表达。结果免疫组化检测慢性肝炎患者、重型肝炎患者及非乙肝对照肝组织G2D阳性细胞百分率分别为13.33%、42.86%和11.11%,差异有统计学意义(χ2=7.95,P〈0.05);重型肝炎患者G2D阳性细胞百分率与慢性肝炎患者比较差异有统计学意义(χ2=5.67,P〈0.05);慢性肝炎患者G2D阳性细胞百分率与非乙肝对照比较差异无统计学意义(χ2=0.05,P)0.05)。流式细胞仪检测重型肝炎患者、慢性肝炎患者及非乙肝对照外周血G2D阳性细胞百分率分别为(12.32±3.43)%、(17.22±5.43)%和(11.11±3.21)%,差异有统计学意义(P〈0.05);慢性肝炎患者G2D阳性细胞百分率与非乙肝对照比较差异无统计学意义(P〉0.05)。结论乙肝患者肝组织及外周血G2D高表达,可能与肝炎的发生相关。
Objective To study G2D receptor expression by NK cells in patients with hepatitis. Methods Subjects were 30 patients with chronic hepatitis B and 21 patients with severe hepatitis were who seen at this hospital from May 2012 to May 2013 as well as 18 healthy individuals. Flow cytometry and immunohistochemistry (streptavidin peroxidase) were used to detect expression of G2D in peripheral blood and liver tissue. Results Immunohistochemistry results indi- cated that patients with chronic hepatitis B, tested positive for G2D at a rate of 13.33 %, patients with severe hepatitis tested positive at a rate of 42.86%, and healthy individuals tested positive at a rate of 11.11%. There were significant differences (χ2 =7.95, P〈0.05) in the rate of positivity for G2D. Liver tissue from patients with severe hepatitis tested positive for G2D at a significantly higher rate than did liver tissue from patients with chronic hepatitis B and healthy indi viduals (2 5.67, P〈0.05). Liver tissue from patients with chronic hepatitis B and healthy individuals did not differ significantly (χ2 =0.05, P〉0.05) in terms of positivity for G2D. Results of flow cytometry indicated that the level of G2D expression was (12.32±3.43)% in peripheral blood from patients with chronic hepatitis B, (17.22±5.43)% in pe- ripheral blood from patients with severe hepatitis, and (11. 11±3.21)% in peripheral blood from healthy individuals. There were significant differences (t= 15.43, P〈0.05) in the level of G2D expression. There were no significant differ- ences (χ2 =0.05, P〉0.05) in the level ofG2D expression in peripheral blood from patients with chronic hepatitis B and in peripheral blood from healthy individuals. Conclusion The level of G2D expression in liver tissue and peripheral blood is related to the development of hepatitis B.
