摘要
目的探讨益气消癥方治疗子宫肌瘤的可能作用机制。方法采用去势+雌激素诱导建立豚鼠子宫肌瘤模型,将68只造模成功豚鼠随机分为模型组(15只)、中药高剂量组(14只)、中药中剂量组(12只)、中药低剂量组(13只)及西药组(14只),并设正常对照组(15只)。去势术后第21日开始,中药高、中、低剂量组分别给予益气消癥方4g/kg、2g/kg、1g/kg干粉;西药组给予米非司酮片1.94mg/kg。各组灌胃容积为1ml/(100g·d),正常对照组及模型组给予等容积蒸馏水,连续12周。检测各组豚鼠子宫组织缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)蛋白表达,并进行HIF-1α与VEGF相关性分析。结果模型组豚鼠子宫组织HIF-1α、VEGF蛋白表达显著高于正常对照组(P<0.05或P<0.01)。与模型组比较,中药高、中剂量组和西药组豚鼠子宫组织HIF-1α、VEGF蛋白表达明显降低(P<0.05或P<0.01);中药高剂量组豚鼠子宫组织HIF-1α蛋白表达显著低于中药中、低剂量组(P<0.01),中药高剂量组豚鼠子宫组织VEGF蛋白表达显著低于中药中、低剂量组和西药组(P<0.01)。HIF-1α与VEGF相关性分析显示,相关系数为0.930,P=0.000。结论益气消癥方可能是通过降低子宫组织HIF-1α、VEGF蛋白表达,抑制血管生成,从而达治疗子宫肌瘤的作用。
Objective To research the possible mechanism of Qi-Boosting Mass-Eliminating Formula for uterine fibroids. Methods The guinea pig models of uterine fibroids were established with castration and estrogen. Totally 68 guinea pig models were randomized into the model group ( n = 15 ), Chinese medicine high-dose group ( n = 14), Chinese medicine middle-dose group ( n = 12) , Chinese medicine low-dose group ( n = 13 ) and western medicine group ( n = 14). The normal control group was established with 15 pigs in it. The Chinese medicine high-dose, mid- dle-dose and low-dose groups were given 4 mg/kg, 2 mg/kg and 1 mg/kg Qi-Boosting Mass-Eliminating Formula drypowder resp Tablets. Th model group were gavaged with the same volume of distilled water. The administration lasted tbr 12 weeks. The protein expressions of hypoxia-inducible factor-1α (HIF-lot) and vascular endothelial growth factor (VEGF) in uterine tissue in each group were observed. The correlation between HIF-1α and VEGF was analyzed. Results The protein expressions of HIF-1α and VEGF in the model group were significantly higher than those in the normal control group ( P 〈0. 05 or P 〈0.01 ).
