离子色谱法测定灵芝孢子粉中多糖的含量 被引量:15

Determination of Monosaccharide Composition of Polysaccharide in Ganoderma lucidum spore by ion Chromatography

摘要 目的建立一种测定灵芝孢子粉中多糖的单糖组成及含量的方法。方法采用沸水浴加热提取灵芝孢子粉中多糖,以2mmol·L^-1三氟乙酸(TFA)水解多糖。采用离子色谱法进行测定,色谱柱为CarboPac PA10(4mm×250mm.10μm);淋洗液:2.5mmol·L^-1 KOH溶液;柱温30℃;流速1.0mL·min^-1。结果9种单糖在一定浓度范围内与色谱峰面积有良好的线性关系,加标回收率为85.6%~97.0%,RSD均小于3%;灵芝孢子粉中多糖主要由葡萄糖、半乳糖、甘露糖、木糖、岩藻糖等单糖组成。结论本实验所建立的方法分离效果好,灵敏度高,可用于灵芝孢子粉中多糖成分的分析研究. OBJECTIVE To develop a method for determination of monosaccharide composition of Ganoderma lucidum spore polysaccharide. METHODS Ganoderma lucidum spore polysaccharide was extracted in boiling water and hydrolyzed by 2 mol·L^-1 TFA. The monosaccharides were determined by ion chromatography on a CarboPac PAIO column, using 2.5 mmol·L^-1 KOH as mobile phase. The column temperature was 30 ℃. The flow rate was 1.0 mmol·min^-1 RESULTS In the certain range, Ihe concentration of each monosaccharide had good linearity with its chromatographic peak area. The recoveries of nnmo- saecharides ranged from 85.6% to 97.0% with RSDs less than 3%. The results showed that Ganoderma lucidum spore polysac- charide was composed of glucose, galactose, mannose, xylose, arabinose, fucose, etc. CONCLUSION The proposed method has good separation effect and high sensitivity, which can be used for the determination of monosaccharide composition of Gamderma lucidum spore polysaccharide.
