摘要
长白山野生灵芝 (Gamoderma lucidum)通过组织分离获得的灵芝 CL988菌株经 60 Co-γ射线与紫外线交替累积诱变获得菌丝高长速灵芝菌株 CJL990 (已经中国科学院微生物研究所鉴定 ) .本文探讨了供试与诱变灵芝菌株的过氧化物酶和酯酶同工酶的电泳分析 ,进一步确定它们的亲缘关系和差异 .结果表明 :(1 )灵芝 CL988菌株和灵芝 CJL990菌株 ,具有灵芝的某些共同的遗传和生物学特性 ,但他们在酶带条数 RF值尚有差异 ,说明诱变灵芝菌株遗传基因有所改变 ;(2 )两种灵芝的菌丝体的 EST和 POD同工酶谱比较稳定 ,完全可以作为鉴定灵芝种类菌属变异的科学依据 .
A strain CJL990, which mycelial growth faster, was induced from the strain CL988 by ~ 60Co-γ ray and UV. The strain CL988 was gained by tissue isolation from the wild Ganoderma lucidum of the Chang Bai Mountain (The strain CJL990 has been identified by microbiology Institute, the Academy of Science of China). The isoenzymes (Esterase, Peroxidase) in the two strains were studied. The results show that there are three more bands of enzyme in CJL990 and differences of RF value between these two strains, that means the genes of the induced strain are changed. Because of its stable, the technique of isoenzyme can be used for identification of Ganoderma lucidum.
