聚ADP核糖基聚合酶的抑制剂苯甲酰胺在肺癌细胞PG凋亡中的作用

摘要 研究了聚 ADP核糖基聚合酶 ( PARP)抑制剂苯甲酰胺 ( BA)在 DNA损伤所造成的肺癌细胞 ( PG)凋亡过程中的作用 .分别用 MNNG( N-甲基 -N′-亚硝基氮亚硝基胍 )和 BA,以及同时用 MNNG和 BA处理 PG细胞 ,然后用非放射性细胞增殖测定试剂盒和流式细胞仪测定 PG细胞的增殖活性和细胞凋亡的变化 ,并用免疫组化法检测细胞中 Bcl-2蛋白表达的变化 .结果表明 ,在 MNNG的作用下细胞的增殖活性受到明显抑制 ,细胞凋亡显著 ,Bcl-2蛋白低表达 .单独用 BA处理 PG细胞时 ,细胞的增殖活性、细胞凋亡数及 Bcl-2的表达与空白对照组相似 .用 BA和 MNNG共同处理 PG细胞时 ,细胞的增殖活性、细胞凋亡数及 Bcl-2的表达 ,与对照组细胞和 BA单独处理的细胞相比均无明显差异 .这说明 PARP对诱导细胞凋亡起重要作用 ,而且这种作用可被 To study the function of poly (ADP ribose) polymerase (PARP) in the apoptosis of lung cancer cells PG caused by DNA damage, the cells were treated by N methyl N' nitro N nitrosogunidine (MNNG), the inhibitor benzamide (BA) of PARP, and the mixture of them, respectively. Non radioactive cell proliferation assay was used to analyze the different proliferation of PG in these cases. The method of FCM was also used to survey the apoptosis cells in PG. Meanwhile, the immunohistochemical method was used to detect the expression of Bcl 2 in PG. The results indicated that under the induction of MNNG, the proliferation of PG was inhibited definitely, PG had definite apoptosis reaction and a low expression of Bcl 2. When PG cells were treated by benzamide alone, their proliferation and apoptosis resemble the control, the expression level of Bcl 2 was as high as the control. When the mixture of benzamide and MNNG was used the proliferation, apoptosis, and expression of Bcl 2 of PG had also no definite differences with those of the control and the BA group. These results show that PARP plays an important role in the induction of PG apoptosis and the function is inhibited by the inhibitor of PARP, benzamide.$$$$
