钙调蛋白依赖性蛋白激酶Ⅱ在AD样大鼠海马的表达及其与细胞凋亡的关系 被引量:6

Expression of Calcium / calmodulin-dependent kinaseⅡ in the hippocampus of Alzheimer's disease-like rats and its relationship with cell apoptosis

摘要 目的研究钙调蛋白依赖性蛋白激酶Ⅱ(CaMKⅡ)在阿尔茨海默病(AD)样大鼠海马的表达及其与细胞凋亡的关系。方法实验大鼠随机分为正常对照组、模型对照组和AD模型组,每组20只。应用Aβ1-40和AlCl3双干预法建立AD样大鼠模型。采用TUNEL染色法检测大鼠海马细胞凋亡情况,采用免疫印迹法检测CaMKⅡ在大鼠海马的表达变化。结果 AD模型组大鼠海马凋亡细胞在海马CA1区、CA3区和DG区均显著升高,与模型对照组和正常对照组相比,差异均有统计学意义(P均<0.05)。AD模型组大鼠海马CaMKⅡ表达升高,与模型对照组和正常对照组相比,差异均有统计学意义(P均<0.05)。结论 CaMKⅡ在AD样大鼠海马表现为病理性高表达,这种病理性高表达对于AD样大鼠海马神经细胞凋亡具有促进作用。 Objective To investigate the expression of Calcium/calmodulin-dependent kinase II ( CaMK II ) in the hip- pocampus and its relationship to the apoptosis of hippocampal neurons in Alzheimer's disease (AD)-like rats. Methods The healthy aged male SD rats were randomly divided into three groups: normal control group, model control group and AD-like model group, with 20 rats in each group. Aβ1-40 and A1C13 were used to establish AD-like rat model. Apoptosis of neurons in the three regions of hippocampus of each group were observed by TUNEL staining and the expressions of CaMK [I in each group were detected by Western blotting. Results Compared with model control group and normal control group, the number of apoptotic cells in AD-like model group were significantly increased in CA1, dentate gyrus and CA3 subfields of the hippocampus ( all P 〈 0.05 ). Compared with model control group and normal control group, the expression of CaMK 11 in AD-like model group was significantly increased in the hippocampus ( all P 〈 0.05 ). Conclusion The expression of CaMK II pathologically increases in the hippocampus of AD-like rat, which promotes the apoptosis of hippocampal neurons.
