低温条件下小鼠树突状细胞功能改变及玉屏风散的调节作用 被引量:2

摘要 目的探讨低温条件下小鼠树突状细胞(DC)功能改变及玉屏风散的调节作用。方法将50只小鼠随机均分为对照组(室温25℃)、模型组(室温10℃)及玉屏风散小、中、大剂量组,给药第4天,将模型组及玉屏风散小、中、大剂量组置于低温容器内,连续低温处理3 d,每天6 h。采用流式细胞术检测各组DC表面分子MHC-Ⅱ、CCR7、CD86表达,RT-PCR法检测CD86、CCR7 mRNA表达。结果与对照组比较,模型组体温下降,MHC-Ⅱ、CCR7、CD86表达降低,CCR7、CD86mRNA表达下降(P均<0.05);与模型组比较,玉屏风散各组的MHC-Ⅱ、CCR7、CD86表达升高(P均<0.05)。结论低温可诱使DC下调MHC-Ⅱ、CD86及CCR7表达,玉屏风散可逆转上述分子表达,增强DC的信息传递功能。 Objective To investigate the changes of the function of mouse dendritic cells under the condition of hypothermia and the regulation of Yu Ping Feng pulvis.Methods Fifty BALB/c mice were randomly divided into 5 groups:the control group at room temperature of 25 ℃,model group at low temperature of 10 ℃,low dose,medium dose and high dose groups of Yu Ping Feng pulvis.And on the fourth day of medication,the other four groups except the control group were placed into the equipment of hypothermia for another three days,and each day for six hours.The expression of MHC class Ⅱ molecules,co-stimulatory molecules CD86 and chemokine receptor CCR7 on dendritic cells was detected by flow cytometry; the levels of CD86,CCR7 mRNA were measured by reverse transcription-polymerase chain reaction (RT-PCR).Results Compared with the control group,the rectal temperature of model group at hypothermia was lower,and so was the expression of MHC class Ⅱ,CCR7,CD86 and the mRNA expression of CCR7,CD86 (all P < 0.05) ; compared with the model group,Yu Ping Feng pulvis enhanced the expression of MHC-Ⅱ,CCR7 and CD86 (all P < 0.05).Conclusions Under the condition of hypothermia,the expression of MHC class Ⅱ,CCR7 and CD86 on the surface of dendritic cells is down-regulated,while the phenomena could be reversed by Yu Ping Feng pulvis and then it enhances the function of information transfer of dendritic cells.
