摘要
目的观察壮精合剂对初老大鼠血清、肝、脑、卵巢中总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量的影响,探讨其抗衰老作用。方法选用12月龄,体重(300±20)g初老大鼠50只,随机分为5组,空白对照组、阳性药对照组、壮精合剂高、中、低剂量组,每组10只。各组分别灌胃相应药物8周后,分光光度法检测大鼠血清、肝、脑、卵巢中T-SOD、GSH-Px、MDA值。结果与空白对照组比较,壮精合剂各组、阳性药对照组大鼠血清、肝、脑、卵巢T-SOD和GSH-Px活性显著升高(P<0.01或P<0.05),MDA含量显著降低(P<0.01);与阳性药对照组比较,壮精合剂高、中剂量组大鼠血清、肝、脑、卵巢T-SOD和GSH-Px活性显著升高,MDA含量显著降低(P<0.01或P<0.05)。结论壮精合剂能明显提高血清、肝、脑、卵巢中T-SOD、GSH-Px活性和降低MDA含量,提高机体对自由基损害的防御能力,增强其抗氧化能力,提示壮精合剂能起到抗衰老的作用。
Objective To observe the effects of Zhuang Jing mixture (Chinese herbal medicine) on the ac- tivities of total superoxide dismutase (T--SOD) and glutathione peroxidase (GSH--Px), the contents of ma- londialdehyde (MDA) in the serum, liver, brain and ovary of the initial aging rats, and to investigate the anti --aging mechanism of Zhuang Jing mixture (ZJM). Methods weight of (300 i- 20) g were randomized to 5 groups, blank groups of ZJM, positive--medicine control group, with 10 rats Fifty initial senile rats at 12 months of age and control group, low--, mid-- and high--dose in each group. After 8 weeks drug administra- tion, a spectrophotometric method was used to detect the contents of T--SOD, GSH--Px, MDA in the liver, brain, ovary and other tissues as well as the serum of them. Results Compared with the blank control group, ZJM of each dose groups and the positive--medicine control group could improve the activities of T-- SOD and GSH--Px ( P 〈0.01 or P 〈0.05) and decrease the contents of MDA( P 〈0.01) in the serum, liver, brain and ovary of aging rats obviously. Compared with the positive--medicine control group, high dose and middle dose of ZJM could improve the activities of T--SOD and GSH--Px and decrease the contents of MDA in the serum, liver, brain and ovary of aging rats obviously ( P 〈0.01 or P〈0.05). Conclusion Zhuang Jing mixture can significantly improve the contents of T--SOD, GSH--Px in the serum,liver, brain and ovary, and lower the level of MDA, and increase the defense capability to free radical damage, and enhance the anti--oxi- dative capacity, which suggest that Zhuang Jing mixture have anti--aging effect.
