利用斑马鱼模型评价川续断皂苷Ⅴ和Ⅵ的抗骨质疏松活性 被引量:18

摘要 为探讨斑马鱼模型评价中药微量成分在体抗骨质疏松活性的适用性与合理性,以25μmol/L泼尼松龙诱导的斑马鱼幼鱼骨质疏松模型考察川续断皂苷V和川续断皂苷VI抗骨质疏松活性。采用茜素红对斑马鱼幼鱼骨骼染色,并以显微检测、数码成像方法定量分析骨骼染色区域。结果 25μmol/L泼尼松龙使斑马鱼骨量显著丢失,与模型组比较,川续断皂苷V和川续断皂苷VI及依替膦酸二钠阳性药均能阻止泼尼松龙诱导的斑马鱼骨量丢失,提示斑马鱼模型成功评价了微量川续断皂苷V和川续断皂苷VI的抗骨质疏松活性,具有在体化、微板化,简单、高效的特点。 To explore the applicability and rationality of zebrafish model for the evaluation of in vivo anti-osteoporosis activity of micro-amount of components from traditional Chinese medicine, zebrafish larvae osteoporosis model induced by 25 μmol/L prednisolone was used to test the anti-osteoporosis activity of asperosaponins V and VI. Zebrafish bones were stained with alizarin red, and quantitative analysis of the stained area was performed by microscopic inspection and digital imaging methods to reflect the amount of zebrafish head skeleton mineralization. The results indicated that 25 μmol/L prednisolone could make zebrafish bone lose significantly, and that compared with the model group, asperosaponins V and VI and etidronate disodium could prevent such loss induced by prednisolone. The results suggested that zebrafish model could be used in evaluating anti-steoporosis activity of micro-amount of asperosaponins V and VI, which has advantages of being in vivo, micro plate, simple and highly efficient.
