摘要
以骆驼蓬为研究对象,选取乙醇浓度、料液比、超声功率、回流时间4个因素进行正交试验,建立了骆驼蓬中总生物碱的提取工艺模式,并测定最佳工艺下提取的骆驼蓬总生物碱对羟自由基(·OH)和氧自由基(·O2-)的清除作用。结果表明,最佳提取工艺为:乙醇浓度55%,料液比为15∶1,超声功率300 W,回流时间为2.0 h,在此最佳工艺下,骆驼蓬总生物碱的提取含量为19.94 mg/g;提取的骆驼蓬总生物碱对·OH和·O2-清除效果显著。
Orthogonal test was carried out to optimize extracting process of total alkaloid from Herba Pegani Harmalae,(TAH ) based on tour factors, namely, ethanol concentration, solid-liquid ratio, uhrasonie power and rcilux time. The extract under optimal conditions was studied for its scavenging activity on hydroxyl radicals (·OH) and oxygen free radical (·O2-). Results showed that the optimal extraction process was as follows: 55% of ethanol concentration, solid-liquid ratio is 15:1, ultrasonic power was 300 W with reflux time of 2.0 h. Under lhe optimum process, the yield of total alkaloid was 19.94 mg/g. Meanwhile, total alkaloid from tterba Pegani Harmalae present remarkable scavenging effect on ·OH and ·O2-.
