摘要
目的探讨前列腺特异性抗原(prostatic specific antigen,PSA)、嗜铬粒蛋白A(chromograninA,CgA)及前列腺癌基因3(prostate cell antigen3,PCA3)的表达在前列腺癌诊断中的价值。方法选择延安大学附属医院2009年1月—2012年1月收治的58例前列腺癌作为前列腺癌组;另选择同期于该院体检的50例无前列腺疾病的健康男性作为对照组。采用化学发光免疫染色法对两组血清PSA进行检测;按法国CIS Biotechnologies international产品试剂盒说明书步骤对2组血清CgA进行检测;按经典总RNA抽屉试剂盒说明书步骤对两组血清PCA3进行检测。分析两组血清PSA、CgA及PCA3的表达水平,并根据Je wett-Whitmore分期标准对前列腺癌组患者进行分期,分析不同分期前列腺癌患者血清PSA、CgA及PCA3的表达水平。结果前列腺癌组血清PSA、CgA、PCA3表达水平显著高于对照组(P<0.01);58例前列腺癌中B期20例,C期13例,D期25例,血清PSA、CgA及PCA3表达水平D期均明显高于B、C期(P<0.01),C期高于B期(P<0.01)。结论 PSA、CgA及PCA3在前列腺癌患者的血清中呈高表达,且表达水平与病情进展呈正相关,可作为前列腺癌诊断的标志物。
Objective To investigate the value of expressions of prostatic specific antigen ( PSA), chromograni- nA (CgA) and prostate cell antigen3 (PCA3) in diagnosis of prostate cancer. Methods A total of 58 patients with prostate cancer in our hospital during January 2009 and January 2012 were selected as the prostate cancer group (group A) ; another 50 healthy people taking medical examinations in our hospital at the same time were chosen as control group. Serum PSA levels in two groups were detected by chemiluminescence immunostaining method; serum CgA 2 levels in the two groups were detected according to CIS Biotechnologies international product kit of manual steps in France; serum PCA3 in the two groups were detected according to the classical total RNA drawer kit of manual steps. Expression levels of serum PSA, CgA and PCA3 in the two groups were analyzed, and the patients in group A were performed stages by the Je wett - Whitmore staging standard. Expression levels of serum PSA, CgA and PCA3 in patients with prostate cancer at different stages were also analyzed. Results Expression levels of serum PSA, CgA and PCA3 leveling group A were sig- nificantly higher than those in control group (P 〈 0. 01 ) ; among the 58 patients in group A, 20 patients in stage B of prostate cancer, 13 in stage C and 25 in stage D, expression levels of serum PSA, CgA and PCA3 of patients in stage D were significantly higher than those of patients in stage B and C (P 〈 0.01 ), and those of patients in stage C were signifi- cantly higher than those of patients in stage B (P 〈 0.01 ). Conclusion Expressions of serum PSA, CgA and PCA3 are high in patients with prostate cancer, and the expression levels are positively correlated with disease progression, which can be used as diagnostic markers of prostate cancer.
