摘要
目的 研究阳离子脂质体介导的小鼠白细胞介素 2基因修饰肝癌细胞瘤苗的抗肿瘤作用。方法 利用阳离子脂质体Transfectam ,将携带小鼠白细胞介素 2 (IL 2 )基因的真核表达质粒VR 1110 ,导入小鼠肝癌细胞H 2 2 ;采用酶联免疫吸附测定法(ELISA )检测转染后经放射线照射、未经放射线照射和冻融后的细胞培养上清液中白细胞介素 2的浓度 ;并研究转染后经放射线照射而制成的瘤苗的抗肿瘤效应。结果 转染后经放射线照射和未经放射线照射的H 2 2细胞均持续分泌IL 2达 30天以上 ;转染细胞在冻融后也能分泌IL 2 ;制备的瘤苗对小鼠肝癌模型肿瘤的生长有明显的抑制作用 ,并能明显提高荷瘤小鼠的生存率。结论 阳离子脂质体介导的小鼠白细胞介素 2基因转染后 ,能在肝癌细胞中有效表达 。
Objective To study the antitumor effect of mouse interleukin 2 gene modified liver cancer vaccines by cationic liposome mediated transfection.Methods Mouse heptocellular carcinoma cell H22 was in vitro transfected an eukaryotic expression plasmid VR1110 encoding the gene for mouse interleukin 2(IL 2) by cationic lipids transfectam.IL 2 concentration was measured from tissue culture supernatants of transfected H22 cells,including those treated with irradiation,without irradiation and with thaw,by enzyme linked immunoabsorbent analysis.Antitumor effect of tumor vaccines composed of irradiated transfection H22 cells was observed.Results IL 2 secretion of H22 cells,either with irradiated or without irradiated,persisted for at least 30 days post transfection and was unaffected by freeze thawing.The growth of tumor in model mice was significantly inhibited after treated with mIL 2 gene modified liver cancer vaccines and the survival rate in model mouse group treated with the vaccines was higher than in control groups( P <0.05).Conclusion Cationic liposome mediated transfection can achieve an efficient mIL 2 gene expression in mouse hepatocellular carcinoma cells H22;The liver cancer vaccines produced by this method can induce potent antitumor effect.
