摘要
目的 探讨p16、p2 1、p5 3、cyclinD1在食管癌中的表达及其作用。 方法 采用免疫组化方法检测 30例食管癌中p16、p2 1、p5 3、cyclinD 1的表达 ,采用PCR SSCP法检测 p5 3基因突变情况。 结果 p16、p2 1阳性表达率均为 6 3 .3 % ,p5 3为6 6 .7% ,cyclinD 1为 86 .7% ,p5 3基因突变率为 2 6 .7%。 结论 食管癌以 p5 3第 7外显子突变为主。cyclinD 1过度表达在食管癌中是常见的并且是早期出现的分子异常 ,p16、p2 1可作为判断食管癌分化程度及预后的 1个参考指标。
Objective To study the expression and the significance of p16,p21,p53,cyclin D1 in esophageal carcinoma.Methods p16,p21,p53 and cyclin D1 were detected by immunochemistry method in 30 cases of esophageal carcinoma and p53 gene mutation was examined by PCR SSCP.Results The positive expression rate of p16,p21 and p53 was 63.3%,63.3% and 66.7% respectively,cyclin D1 expression rate was 86.7%;the p53 gene mutation rate was 26.7%.Conclusion The mutation of exon 7th of p53 gene is most commonly seen in these cases of esophageal carcinoma.The overexpression of cyclin D1 is often seen in early cases.p16 and p21 may be as an associated factor to estimate the degree of differentiation and prognosis of esophageal carcinoma.
