摘要
目的 观察桂皮酸 (CINN)对SMMC 772 1细胞的诱导分化作用。方法 体外培养人肝癌细胞SMMC 772 1,加入不同浓度的CINN ,通过对其形态学、细胞增殖动力学、细胞生物化学行为及细胞周期分布的观察 ,探讨CINN对肿瘤细胞的诱导分化能力。结果 经CINN处理后 ,SMMC 772 1细胞形态趋向良性分化 ,生长减速慢 ,集落形成率显著降低 ,甲胎蛋白分泌减少 ,FCM检测显示G0 /G1期细胞增加而G2 +M期细胞减少。结论 CINN对SMMC 772
Objectives To study the induced differentiation effect of Cinnamic Acid on Human Hepatocarcinoma Cell.Methods Human hepatocarcinoma cells(SMMC 7721) were cultured and treated with 1 6mmol/L cinnamic acid(CINN) in vitro and their induced differentiation effect in terms of morphology,proliferation,colony forming ability on semisolid agar culture,biochemical behavior and cell cycle changes were observed.Results These results showed that CINN could inhibit proliferation of SMMC 7721 cells.The secretion of α fetoprotein(AFP) was significantly lower than that of the control cells.Flow cytometry(FCM) demonstrated that G2+M phase cells decreased and G0/G1 Phase cells increased.Conclusion CINN could have the effect of induced differentiation on SMMC 7721 cells.We propose that CINN should be further evaluated in preclinical and clinical studies,in pursuit of the development of alternative approaches to hepatocarcinoma management.
