RGS17在不同肝癌细胞株及HuH7裸鼠移植瘤中的表达

摘要 目的研究G蛋白信号转导调节因子17(RGS 17)在人肝癌细胞株和正常肝细胞株中的表达差异以及在皮下成瘤动物模型中的表达。方法从2种肝癌细胞株HepG2、HuH7及正常肝细胞株THLE-2中提取总RNA,用逆转录酶将总RNA逆转成cDNA,以cDNA为模板,应用实时荧光定量聚合酶链反应法检测RGS17 mRNA表达水平;在裸鼠皮下接种肝癌细胞株HuH7,成瘤后检测瘤组织与肝组织中RGS17 mRNA的表达差异。结果 (1)在肝癌细胞株HepG2、HuH7以及正常肝细胞株THLE-2中均检测到RGS17 mRNA的表达;(2)与正常肝细胞株THLE-2相比,肝癌细胞株HepG2、HuH7中RGS17 mRNA的表达明显增高,差异有统计学意义(P<0.05);(3)在HuH7肝癌细胞株裸鼠成瘤模型的瘤块组织中检测到RGS17 mRNA表达,且与肝组织相比,瘤块组织中RGS17 mRNA表达显著增高,差异有统计学意义(P<0.05)。结论体外细胞株和裸鼠动物体内2个层次的研究结果表明,在肝癌细胞中RGS17 mRNA表达上调,提示RGS17可能参与了肿瘤的发生和发展,为肝癌发病机制的研究提供了新思路,并为肝癌的预防和治疗提供了新的分子靶点。 Objective To study the differential expression of regulator of G protein signaling 17 (RGS 17) between hepatocellular carcinoma cell lines and normal liver cell lines of human and the expression in subcutaneous animal model of tumor. Methods Total RNA was extracted in hepatocelluar carcinorma cell lines HepG2, HuH7 and normal liver cell line THLE-2. Reverse transcrip tase was used to obtain template cDNA by reversing the total RNA into cDNA,and the expression of mRNA of RGS17 was detected by real-time fluorescent quantitative polymerase chain reaction (RT-PCR). Hepatoma cel line HuH7 was injected subcutaneously in nude mouse, the differential expressions of mRNA of RGS17 in tumor tissue and liver tissue were measured after tumor formation. Results ( 1 ) RGS17 mRNA expression was detected in hepotocarcinoma cell lines HepG2, HuH7 and normal liver cell line THLE-2. (2)Compared with normal liver cell line THLE-2,the RGS17 mRNA levels in HepG2 and HuH7 were obviously higher with statistically significant difference (P〈0.05). (3)The RGS17 mRNA expression was detected in nude mouse tumour, and the expression of RGS17 mRNA in nude mouse tumor tissue was elevated significantly than that in the liver tissue with statistically significant difference (P〈0.05). Conclusion The research results from in vitro and in vivo cell lines show that RGS17 mRNA is up-regulated in the hepatocelluar carcinoma cells ,which suggests that RGS17 might play a role in the occurrence and development of tumor. The results provide a new method for the study of pathogenesis of hepatocellular carcinoma, and a molecular target for the prevention and treatment of hepatocelluar carcinoma.
