江西部分地区303株结核分枝杆菌基因分型研究 被引量:2

Study on genotyping of 303 mycobacterium tuberculosis isolates in some areas of Jiangxi province

摘要 目的选取2008年-2009年间由南昌市疾病预防控制中心收集的江西省部分地区结核分枝杆菌临床分离株,通过多位点可变数目串联重复序列分型,了解南昌市结核分枝杆菌流行菌株基因分型情况。方法对临床分离株进行常规培养,收集菌体,提取基因组DNA,采用聚合酶链反应(PCR)分别对15个VNTR位点进行检测分析,基因聚类分析采用BioNumeics软件。结果经聚类分析,303株结核分枝杆菌分为4个基因群(I群、Ⅱ群、Ⅲ群、Ⅳ群)119个基因型。聚类分析得知,Ⅰ群占86.47%,可分成91个基因型;Ⅱ群占3.96%,可分成6个基因型;Ⅲ群占4.95%,可分成11个基因型;Ⅳ群占4.62%,可分成11个基因型。结论江西部分地区的结核分枝杆菌存在基因多态性,其主要流行型为Ⅰ型。 Objective To analyze the genotypes of epidemic mycobacterium tuberculosis strains in Nanchang by multiple - locus variable number tandem repeat analysis (MLVA) in some areas of Jiangxi province from 2008 to 2009. Methods M. tuberculo- sis were cultured on Lowenstein -Jensen medium to extract the bacteria genome DNA. Then polymerase chain reaction(PCR) was used to detect the VNTR locus in the DNA samples. The clustering analysis of genotypes was done with BioNumerics. Results These M. Tuberculosis strains could be categorized into 4 gene clusters( I, II, llI, IV) consisting of 119 genotypes. Clus- ter I included 91 genotypes, accounting for 86.47%, cluster 1I included 6 genotypes, accounting for 3.96% , cluster 11I included 11 genotypes , accounting for 4.95%, cluster IV included 11 genotypes, accounting for 4. 62%. Conclusion M. tuberculosis showed obvious gene polymorphisms in part area of Jiangxi province, and cluster I was the predominant prevalent strain.
