摘要
目的:探索CD44v10剪接变异体对鼻咽癌细胞增殖的生物学作用。方法:将CD44v10基因编码序列重组,构建pIRESneo3-CD44v10重组质粒表达载体,重组表达载体扩增后采用脂质体转入鼻咽癌6-10B细胞系,G418筛选稳定转染细胞株;RT-PCR和蛋白印迹鉴定CD44v10重组表达6-10B细胞株;MTT法分析细胞增殖。结果:获得了CD44v10稳定重组表达的6-10B细胞株;CD44v10表达抑制6-10B细胞增殖。结论:CD44v10表达对鼻咽癌细胞的恶性增殖可能具有抑制作用。
Objective: To explore the biological function of splice variants of CD44v10 on the proliferation of nasopharyngeal carcinoma cells. Methods: The sequence of CD44v10 gene was constructed with pIRESneo3 plasmid expression vector to build recombinant pIRESneo3-CD44v10, the recombinant expression vector was amplified and transfected into nasopharyngeal carcinoma 6-10B cell line by liposome, the stably transfected cell strains were screened by G418, CD44v10 recombinant expression 6-10B cells were detected by RT-PCR and western blot. Cell proliferation was tested by MTT experiment. Results: The stable CD44v10 expression 6-10B cells were gotten. Expression of CD44v10 inhibited proliferation of 6-10B cells. Conclusion. The expression of CD44v10 may inhibit the malignant proliferation of nasopharyngeal carcinoma cells.
