胆管癌发生发展中FBW7、c-myc及miR-27a的表达及意义 被引量:1

摘要 目的探讨胆管癌发生发展中FBW7、c-myc及miR-27a的表达及意义。方法培养人胆管癌QBC939细胞,用miR-27a siRNA慢病毒转染胆管癌QBC939细胞,转染24 h后,采用Northern blot和相对荧光素酶活性检测FBW7、c-myc及miR-27a的表达。结果 miR-27a siRNA慢病毒成功转染胆管癌QBC939细胞,Northern blot结果显示FBW7表达明显升高,C-myc表达下降。转染了miR-27a siRNA慢病毒的QBC939细胞系,荧光素酶活性明显升高。结论 miR-27a/FBW7/c-myc信号通路在胆管癌的发生发展中能起到重要作用。 [Objective] To study the expression relationship between FBW7, c-myc and miR-27a in human cholangiocarcinoma. [Methods] Cultured human bile duct carcinoma QBC939 gene, and transfected miR-27a siR- NA lentivirus into cholangiocarcinoma QBC939 cells. After 24 hours transfection, used the Northern blot and relative luciferase activity detection to detect the expression of c-myc, miR-27 and FBW 7. [Results] MiR-27a siRNA lentivirus infectd bile duct carcinoma cell line QBC939 successfully. Northern blot results showed that FBW7 ex- pression was significantly increased, C-myc expression decreased. QBC939 cell line transfected with miR-27a siR- NA slow virus increased the lueiferase activity significantly. [ Conclusion ] The miR-27a/FBW7/c-mye pathway has the relationship with the progression of human cholangiocarcinoma.
