摘要
目的 用好氧性培养基和 Hungte压氧操作技术相结合的方法从水稻田土壤中分离三个甲烷氧化菌分离物 ,分别命名为 MOB0 0 4、MOB0 0 7、MOB0 0 9。方法 包括分离培养基的制作 ,生理生化实验 ,CO2 和生长光密度值的测定。结果 三个甲烷氧化菌分离物的革兰染色均为阴性 ,适宜生长 p H为 6 ,范围为 5~ 8,适宜生长温度为 30~35℃。 MOB0 0 9对 p H值和温度具有较宽的适宜范围。结论 试验结果亦表明甲烷氧化菌的良好生长必须有甲烷和氧同时存在 ,都只能利用甲烷、甲酸、甲醇等简单甲基物 ,不能利用其他复杂有机物。其确切的分类地位有待于进一步研究。
Objective To isolate methane oxidizing bacteria from earth to study their characteristics.Methods Combined method of aerobic media and anaerobic operation technique was used for bacterial isolation.Their general characteristics was also studied.Results All three isolates were Gram negative staining.They could just utilize simple methyl compounds such as methane,formate and methanol,etc.,but not other complex organic compounds with methane group.Conclusion It seems that the existence of methane and oxygen at the same time is necessary for optimal growth of methane oxidizing bacteria in the earth.
