RP-HPLC法分析芡种壳提取物中单体酚组成 被引量:1

Analysis of the phenolic compounds in extracts of the euryale ferox seed shell by RP-HPLC method

摘要 建立了一种反相高效液相色谱法分离10种单体酚的方法,并用于芡种壳提取物中单体酚含量分析。采用C18反相色谱柱、紫外检测器、以V(甲醇):V(1%乙酸)=3:7为流动相,在280nm下,可同时分离10种单体酚物质,加样回收率95.3%-101.3%,相对标准偏差在2.1%以下,回归方程在2.5~125mg/L范围内线性良好。方法可分离芡种壳提取物酸处理产物中的单体酚以及绿茶50%乙醇提取物中的单体酚。结果显示,芡种壳提取物中富含芦丁、没食子酸和儿茶素类物质。经适度酸处理后,芡种壳提取物A,B,C和D中的芦丁含量呈数倍增加,从72.6,22.8,22.4和9.2mg/g分别增至297.7,387.2,225.9和141.4mg/g。 A reversed phase-high performance liquid chromatographic method was established to simultaneously determine 10 kinds of phenolic compounds, and the method was used to determine the phenolic compounds in the extracts from the eurgaleferox seed shell (EFSS). A Cls reversed-phase column was utilized for chromatographic separation with a UV detector. Methanol - 1% acetic acid (30:70, V/V) was used as the mobile phase and detection wavelength was at 280 nm, 10 kinds of phenolic references can be simultaneously separated and determined with the average recoveries ranging from 95.3% to 101.3% and the RSDs were less than 2. 1%. The calibration curves have good linearities in the range of 2. 5 - 125 mg/L. In the sample analysis, the major phenolic compounds in 4 kinds of acid hydrolysed EF seed shell extracts( A, B, C and D) and green tea extract (TE) were analyzed. The results indicated that higher content rutin, gallic acid and catechin compounds were detected in the EFSS extracts and its acid hydrolyses. Especially rutin, the contents in A, B, C and D increased several times after acid treatment, from 72.6,22. 8,22.4 and 9.2 mg/g to 297.7,387.2,225.9 and 141.4 mg/ g, respectively. The results suggested that the euryaleferox seed shell is a potential natural resource of turin. The rutin concentration in EFSS extracts increased significantly after treated with proper acid.
