摘要
建立了基于质谱的人肝移植后血清中差异表达蛋白质的分析方法,肝移植前后的人血清蛋白质用双向凝胶电泳分离,AgNO,染色,差异蛋白质斑点用胰蛋白酶进行胶内酶解后,采用纳升超高效液相色谱-电喷雾串联质谱进行鉴定。鉴定了8个显著差异表达的蛋白质,Mascot检索分值101~1543分,肽段匹配数5~34个,序列覆盖率10%~43%。所发现的差异蛋白质多与免疫调节有关,可为深入研究肝移植术后免疫排斥的分子机理提供线索。
In this report the mass spectrometry-based method was established for analysis of differentially expressed protein in human serum after liver transplantation. The proteins in serum before and after liver transplantation were separated by two-dimensional gel electrophoresis with silver staining, and the differevtial proteins on the gel were done in gel digestion with trypsin and then identified by nanoUPLC-ESI-MS/MS.
