摘要
本实验利用差异显示反转录PCR(DDRT PCR)技术分析单眼视觉剥夺大鼠左、右侧视皮质基因差异表达情况 ,并通过反Northern杂交去除假阳性筛选差异表达基因片段。最终获得 3个差异表达基因片段。通过克隆、测序和同源性比较发现 ,其中一个片段 5’端的 166个碱基与人KIAA0 541蛋白的mRNA编码区的166bp区段有 83%的同源性。
Differential display reverse transcription polymerase chain reaction (DDRT PCR) was carried out to analyze the differential gene expression between right and left visual cortex of monocularly deprived rat. The positive differentially expressed fragment was screened out through reverse Northern assay. Three differentially expressed fragments were obtained. After cloning, sequencing and homology comparing, one fragment was found to be a novel EST and showed 83% homology with a 166bp region of homo sapiens KIAA0541 protein mRNA. The present result indicates that monocular vision deprivation induces alternations of gene expression in the visual cortex.
