储存磷光质屏放射自显影在测定兔血浆中^(125)I-重组人尿激酶原浓度的应用 被引量:3

摘要 目的 探讨储存磷光质屏放射自显影结合聚丙烯酰胺凝胶电泳测定兔血中12 5I 重组人尿激酶原 (12 5I rhpro UK)浓度的方法。方法 将含12 5I rhpro UK样品的聚丙烯酰胺凝胶贴于储存磷光质屏上曝光 ,根据储存磷光质屏上检出的吸收度值计算12 5I rhpro UK浓度。结果与结论 该方法特异性、线性及线性范围、灵敏度、重现性、回收率均可达到蛋白多肽药物药代动力学研究的要求。并测定了 0 .8mg·kg-1静脉给药时 (12 5I rhpro UK/rhpro UK =1∶9混合 ) ,兔血浆12 5I rhpro UK浓度 Aim The method of assaying 125 I rhpro UK concentration of plasma in rabbits with combination of storage phosphor screen autoradiography and PAGE were evaluated in this paper.Methods The 125 I rhpro UK concentrations of plasma were calculated according to optical density on the storage phosphor screen exposed to polyacrylamide gel containing 125 I rhpro UK.Results and Conclusion The specificity,linear range, reproducibility and recovery for the method could meet the requirements for studying pharmacokinetics of bioactive proteins or peptides. 125 I rhpro UK concentration time curve of plasma in rabbits was measured when giving 0.8mg·kg -1 (mixture of 125 I rhpro UK/I rhpro UK=1∶9) in vein.
