摘要
采用响应面分析方法优化铁皮石斛原球茎富硒条件,首选利用Plackett-Burman试验进行因素的显著性分析,筛选出具有显著效应的3个因素:硒浓度>培养时间>光照强度;再通过最陡爬坡试验逼近其最大响应区域,最后通过BoxBehnken试验对其进行优化,得到最佳条件为硒浓度、培养时间、光照强度分别为0.31mg/L、58.87d、1 834.09lx,原球茎硒含量预测值为4.72mg/kg。
Response surface methodology (RSM) was used to optimize culture conditions of selenium-enriched of Dendrobii candidum protocorm. Plackett-Burman design assay was used to evaluate the significance of these factors, and three factors including selenium concentrations, culture time and illumination intensity had statistically signifi- cant effects. Then steepest ascent procedures were applied to define optimal response region of these three factors. Finally the optimal factors were determined by adopting RSM analysis. The results showed that selenium concentra- tion was 0.3 lmg/L, culture time was 58.87d and illumination intensity was 1 834. 091x. The predicted value of se- lenium was 4. 72mg/kg.
