摘要
[目的]优化白扁豆多糖的提取条件。[方法]以白扁豆为材料,在单因素的基础上,通过Box-Behnken中心组合设计,选定提取温度、提取时间及料液比为3因素,分别选3个水平进行中心组合试验设计,通过响应面分析,拟合数学模型,建立1种最优的多糖提取方法。[结果]计算得数学模型公式为:Y=0.75+0.11A+0.048B+0.068C-0.040AB+0.050AC+0.045BC-0.046A2+0.034B2-0.061C2;最佳提取条件为:料液比1∶70 g/ml,提取温度为70℃,提取时间为100 min;在此条件下,白扁豆多糖产率为0.96%,3次平均试验所得结果为0.94%。[结论]该优化方法合理、可行。
[ Objective ] To optimize the extraction conditions of polysaccharide from Dolichos lablab L. [ Method ] On the basis of single - factor experiments, the Box - Behnken center - united design principles were used to design the experiments and the response surfaces analysis of 3 fac- tors, including extracting temperature, extraction time and ratio of liquid to solid when deposited polysaccharides, and 3 levels were adopted re- spectively. [ Result ] The mathematical model was obtained: Y = 0. 75 + 0. 11A + 0. 048B + 0. 068C -0. 0aOAB +10. 050AC + 0. 045BC -0. 046A2 + 0. 034B2 -0. 061C2 ; the optimum extraction condition were ratio of liquid to solid 1:70, extraction temperature 70 ℃ and extraction time 100 min, under these condition the yield of polysaccharide from Dolichos lablab L. is 0.96%. Experiments were carried out at these optimum condi- tions, the average yield of polysaccharide is 0.94%. [ Conclusion' These results indicated that this method is feasible and reasonable.
