摘要
目的研究生长分化因子-15(GDF-15)基因多态位点+157A/T:rs1059369与不稳定型心绞痛的相关性。方法临床诊断为冠心病不稳定型心绞痛并冠状动脉造影阳性组50例,对照组冠状动脉造影阴性40例。采用连接酶检测法(LDR)方法对全部病人GDF-15基因多态位点+157A/T进行单核苷酸多态性(SNP)分型检测分析。设计特异上游引物和下游引物,多重PRC扩增SNP,来获得含有突变的目的基因片段,在目的基因片段设定荧光探针,当探针与目的基因片段DNA碱基互补配对后,通过连接酶催化磷酸二酯键将相邻两根探针连接从而形成产物,当探针的两条寡聚核苷酸接头处存在碱基错配,则连接反应便不能进行。最后,通过测序仪电泳读取rs1059369多态分型检测结果。结果①冠状动脉造影阳性者与冠状动脉造影阴性者血清+157A/T:rs1059369[A/T]多态分型两组间不同表型的比较,差异无统计学意义(P=0.57)。②冠状动脉造影阳性者者与冠状动脉造影阴性者血清+157A/T:rs1059369[A/T]多态分型两组间各表型分析,差异无显著性(AT组P=0.63,TT组P=0.45,AA组P=0.59,OO组P=0.26)。结论GDF-15基因多态位点+157A/T:rs1059369的多态性与不稳定型心绞痛无明显相关性。
Objective To investigate the correlation between growth differentiation factor - 15 ( GDF - 15) gene polymorphism loci + 157 A/T and unstable angina pectoris. Methods Single nucleotide polymorphism(SNP) genotyping for GDF - 15 gene polymorphism loci + 157A/T was performed using multiplex PCR and ligase detection reaction (LDR) in 50 patients with unstable angina pectoris and positive coronary angiog- raphy, and 40 coronary angiography negative patients. The test results were read by electrophoresis sequencing instrument. Results ① No statisti- cally significant difference was found between coronary angiography positive patients and negative patients in + 157 A/T rs1059369 [ A/T] poly- morphism phenotypes ( P = O. 57 ). ② No statistically significant difference was found between coronary angiography positive patients and negative patients in each genotypic group of +157 A/T: rs1059369 [A/T] (AT groupP =0.63, TY groupP =0.45, AA groupP =0.59, OO GroupP = 0.26). Conclusion No significant correlation is found between GDF - 15 + 157 A/T: rs1059369 polymorphism and unstable angina pectoris.
