摘要
目的初步探讨1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)帕金森病小鼠模型十二指肠和远端结肠细菌内毒素受体Toll样受体4(TLR4)表达的变化。方法成年雄性C57BL/6小鼠随机分为4组:PBS对照3d组,PBS对照7d组,MPTP帕金森病小鼠3d组,MPTP帕金森病小鼠7d组,MPTP帕金森病小鼠模型组给予MPTP(20mg/kg,3次,每次间隔2h),PBS对照组给予等量PBS。在末次处理后3d或7d处死小鼠,取出十二指肠和远端结肠组织。采用免疫印迹法检测TLR4的表达,采用免疫组化法检测TLR4阳性细胞表达。结果在MPTP帕金森病小鼠7d组,十二指肠、远端结肠TLR4蛋白表达均显著增加(均P<0.05)。结论 MPTP帕金森病小鼠模型十二指肠和远端结肠TLR4上调可能与帕金森病的病理生理进程相关。
Objective To investigate the change of the expression of TLR4 in the duodenum and distal colon of mice with MPTP-induced Parkinson's disease. Methods Adult C57BL/6 mice were randomly divided into four groups:two normal con trol groups(3,7 days)and two MPTP groups(3,7 days). Mice were sacrificed 3 or 7 days after PBS or MPTP administration Eintraperitoneal injection of PBS or MPTP(20 mg/kg,3 times at 2 h apart)]. The expression levels of TLR4 in the duodenmn and distal colon were detected by Western blot and immunohistochemistry. Results The TLR4 expression was significantly ele vated in the duodenum and distal colon 7 days after MPTP treatment, significantly higher than that in the normal control groups (P〈0.05). Conclusion The upregulation of TLR4 in specific areas of the intestine may be associated with the pathophysiology of MPTP-induced Parkinson's disease.
