摘要
目的研究补益营卫方对衰老人皮肤成纤维细胞TGFβ1及其受体基因表达的影响。方法通过传代培养复制衰老人皮肤成纤维细胞模型,利用荧光定量PCR法检测补益营卫方对衰老人皮肤成纤维细胞TGFβ1及其受体基因表达。结果人皮肤成纤维细胞衰老时TGFβ1基因的表达水平相对于年轻细胞明显下降,24h和48h分别下降了69%和44%。补益营卫药物处理后衰老人皮肤成纤维细胞TGFβ1基因的表达明显增加,24,48和72h分别是衰老人皮肤成纤维细胞的7.16,3和2.6倍。人皮肤成纤维细胞衰老时TβI基因表达水平,24h和48h时分别比年轻人皮肤成纤维细胞下降了17.4%和72%,72h时则增加了70%。补益营卫药物处理后TβI基因的表达水平相对于衰老人皮肤成纤维细胞,其24h和48h分别提高了9%、38%,72h时则下降了55%。人皮肤成纤维细胞衰老时TβII基因的表达水平,24h和72h时,衰老人皮肤成纤维细胞分别是年轻人皮肤成纤维细胞的39%和68%,分别下降了61%和32%,而48h时反而比年轻人皮肤成纤维细胞增加了33%。补益营卫药物处理后显著提高了TβII基因的表达,相对于衰老组,其24,48和72h分别提高了2.09,1.37和1.75倍。结论补益营卫方对衰老人皮肤成纤维细胞TGFβ1和TβII基因表达具有促进作用。对TβI受体基因表达具有双向调节作用。
Objective To research the effect of Recipe of Supplementing Ying - wei on the gene expression of TGFβ1 and its receptors in the aging human skin fibroblasts. Methods Copied human skin fibroblasts aging model by cultured cells, to detect the effect of Recipe of Supplementing Ying - wei on the expression of TGFβ1 and its receptors in the aging human skin fibroblasts by using RT - PCR. Results The results showed that the expression of TGF[31 in the aging human skin fibroblasts was significantly declined,24h and 48h respectively decreased by 69% and 44%. After treated with Recipe of Supplementing Ying - wei, the ex- pression of TGF^I increased significantly,24h,48h and 72h was respectively 7.16 times,3 times and 2.6 times of the aging human skin fibroblasts. Compared with the young skin fibroblasts, the expression of Tβ1 in the aging human skin fibroblasts was de- clined,24h and 48h respectively decreased by 69% and 44% ,72h increased by 70%. After being treated with Recipe of Supple- menting Ying -wei, the expression of Tβ1 increased, 24h and 48h respectively increased by 9% and 38%, 72 h declined by 55%. Compared with the young skin fibroblasts, the expression of TβII in the aging human skin fibroblasts was declined,24 h and 72h respectively declined by 61 % and 32% ,48h increased by 33%. After being treated with Recipe of Supplementing Ying - wei, the expression of TβII in the aging human skin fibroblasts increased significantly ,24 h,48 h and 72h wasrespectively 2.09 times, 1.37 times and 1.75 times of the aging human skin fibro - blasts. Conclusion Recipe of Supplementing Ying - wei could promote the expression of TGF^31 and T[3II in the aging of human skin fibroblasts, and also had dual - directional regulation on the expres- sion of the receptor Tβ1.
