加味楂曲饮防治实验性非酒精性脂肪肝药效学及其部分机制研究 被引量:2

Study about the Pharmacodynamics and Its Part Action Mechanism of Jiawei ZhaQuYin Interventing the Experimental Non-alcoholic Fatty Liver Disease

摘要 目的研究加味楂曲饮(JWZQY)干预非酒精性脂肪肝(Non-Alcoholic Fatty Liver Disease,NAFLD)的药效学及部分作用机制。方法 SDF级大鼠随机分为6组:空白组、模型组、对照组、低剂量组、中剂量组、高剂量组。高脂饲料造模,对照组给予灌服等体积(0.038g/ml)复方蛋氨酸胆碱溶液,低、中、高剂量组分别灌服等体积含1,2,4 g/ml浓度JWZQY溶液,常规油红O法观察肝脏组织形态学改变,全自动生化仪测定血清丙氨酸基转移酶(Alanine Transaminase,ALT)、谷氨酸基转移酶(Aspartate Amino Transferase,AST)、甘油三酯(Triglycerides,TG)、总胆固醇(Total Cholesterol,TC)水平,Elisa检测血清脂联素(Adiponection,ADP)、肿瘤坏死因子-α(Tumor Necrosis Factor-alpha,TNF-α),比色法检测血清游离脂肪酸(Free Fatty Acid,FFA)水平。结果高、中剂量JWZQY治疗后大鼠肝脏脂肪变程度明显减轻(P<0.05);与模型组相比,阳性组及高、中剂量组大鼠血清中ALT、AST、TC、TG水平降低,差异有统计学意义(P<0.01)。与阳性组比较,高剂量大鼠血清中ALT、AST、TC、TG水平降低,差异有统计学意义(P<0.01)。与模型组相比,中剂量组ADP、TNF-α、FFA差异有统计学意义(P<0.01)。结论 JWZQY可有效干预NAFLD,推测可能与其改善大鼠胰岛素抵抗(Insulin resistance,IR)、减少FFA生成有关。 Objective To study the pharmaeodynamics and its part action mechanism of JiaWei ZhaQuYin(JWZQY) interventing the Experimental Non - Alcoholic Fatty Liver Disease. Methods SDF rats were randomly divided into six groups: blank group, model group, positive control group, low dose group, middle dose group and high dose group. Rats were given high fat diet to model- ing, the positive control group was given the same volume of compound methionine choline solution(0. 038g/ml) orally. Low, me- dium and high close group was given the same volume of solution containing lg/ml,2g/ml,4g/ml JWZQY accordingly. The con- ventional oil red O method was used to observe the morphological changes of the liver tissue. The automatic biochemical analyzer used to measure the level of serum Alanine transaminase, Aspartate Amino Transferase, Triglyceride, Total Cholesterol, Elisa was used to detect the levels of serum Adiponection, tumor NecrosisFactor - alpha, Colorimetly was used to detect serum Free Fatty Acid. Results High dose and medium dose JWZQY significantly reduced hepatic steatosis (P 〈 0.05 ). Compared with the model group ,The level of serum ALT, AST ,TC ,TG in the control, medium and high dose groups was reduced, the difference was statisti- cally significant (P 〈0.01 ). Compared with the control group, serum ALT, AST, TC, TG of high dose group was reduced, the difference was statistically significant (P 〈 0.01 ). Compared with model group, the difference of serum ADP, TNF -alpha, FFA in medium dose group was statistically significant ( P 〈 0.01 ). Conclusion JWZQY can effectively intervene in the experimental Non -alcoholic Fatty Liver Disease, suggesting that it may be related to improving IR, reducing FFA generate.
