摘要
目的 :研究雷公藤多甙和 IL - 10对人树突状细胞 (DC)表面人类白细胞抗原 - DR(HL A- DR)和 CD80表达及 IL - 12 p40转录和分泌的影响。方法 :通过粒细胞 -巨噬细胞型集落刺激因子 (GM- CSF)、IL- 4和肿瘤坏死因子 α(TNF- α)体外培养体系 ,从人外周血单个核细胞中获得 DC,细胞表面 HL A- DR和 CD80表达采用流式细胞仪分析 ,IL - 12 p40转录和分泌分别采用逆转录多聚酶链反应 (RT- PCR)技术和 EL ISA法。结果 :雷公藤多甙 (5~ 2 0μg/ ml)、IL - 10 (5 0~ 2 0 0 ng/ ml)能下调 DC表面 HL A- DR和 CD80的表达 ,同时雷公藤多甙、IL- 10能抑制 DC内 IL- 12 p40 m RNA的转录和分泌。结论 :雷公藤多甙、IL- 10能通过抑制表面分子的表达和细胞因子的合成而干扰 DC的成熟和功能。
Objective:To investigate the effects of tripterygium wilfordii (T Ⅱ) and interleukin 10(IL 10) on the expressions of HLA DR and CD80 and the production and transcription of IL 12 p40 subunit in human dendritic cells (DC). Methods:DC were generated by culturing peripheral blood mononuclear cell of healthy volunteers with GM CSF, IL 4 and TNF α in vitro . The expressions of HLA DR and CD80 were examined by flow cytometry after the cells being stained with immunofluorescence antibodies. Reverse transcriptase PCR analysis and ELISA were used to detect IL 12 p40 mRNA transcription and protein level, respectively. Results:T Ⅱ 5 20 μg/ml and IL 10 50 200 ng/ml significantly down regulated the membrane expressions of HLA DR and CD80 on DC in a dose dependent manner. IL 12p40 production and mRNA transcription were also inhibited in DC by treating of T Ⅱ and IL 10. Conclusion:T Ⅱ and IL 10 interfere with the maturation and function of human DC by down regulating membrane expressions of HLA DR and CD80 and inhibiting transcription and synthesis of IL 12p40 subunit.
