P38MAPK在大鼠心肌缺血再灌注中HSP70的调控作用机制 被引量:1

摘要 目的探讨P38MAPK在缺血再灌注损伤中的作用,以及与HSP70存在的可能相关关系。方法将40只雄性大鼠随机分为假手术组、模型组、IPC组、阻断剂组四组,通过结扎冠状动脉左前降支建造大鼠心肌缺血再灌注损伤模型,并观察大鼠心肌P38MAPK、HSP70蛋白以及细胞凋亡率的变化。结果与假手术组比较,模型组、IPC组、阻断剂组三组心肌细胞凋亡及P38MAPK、HSP70蛋白表达均明显升高,差异有统计学意义(P〈0.01,P〈0.05);而与模型组对照比较,IPC组心肌细胞凋亡率明显下降,P38MAPK、HSPT0蛋白表达均明显升高,差异均有统计学意义(P〈0.01、P〈0.05),模型组与阻断剂组对照心肌细胞凋亡率及P38MAPK、HSP70蛋白表达均无明显变化。结论P38MAPK信号通路的活化能有效抑制心肌细胞凋亡,改善心肌缺血再灌注损伤,其作用机制可能与热休克蛋白70的激活相关。
