The expression of RECK and MMP-9 protein in oral squamous cell carcinoma and its clinical significance

摘要 目的:研究抑癌基因RECK及基质金属蛋白酶-9(MMP-9)在口腔鳞癌中的表达及临床意义。方法:采用免疫组化SP法检测RECK基因和MMP-9蛋白在60例口腔鳞癌及10例正常口腔黏膜组织中的表达,并分析与临床病理特征的关系。结果:①RECK蛋白在口腔鳞癌组织中的阳性表达率为51.67%,在正常口腔黏膜组织中的阳性表达率为90%,二者差异有统计学意义(P<0.05);②MMP-9在口腔鳞癌中的阳性表达率为78.33%,在正常口腔黏膜组织中的阳性表达率为20%,二者差异也有统计学意义(P<0.05);③RECK和MMP-9蛋白表达呈负相关关系(r=-3.266,P<0.05)。④RECK和MMP-9蛋白在口腔鳞癌中的表达与年龄、性别、肿瘤的大小等临床病理特征无关(P>0.05),而与病理分级、TNM分期及淋巴结转移有关(P<0.05)。结论:RECK及MMP-9的异常表达在口腔鳞癌的发生及发展中起重要作用。RECK展因可能是通过下调MMP-9蛋白的表达水平达到抑制口腔鳞癌的侵袭与转移。 Objective:To investigate the expression of reversion inducing cysteine rich protein with kazal motifs (RECK) and matrix metalloproteinase-9 (MMP-9) in oral squamous cell carcinoma and its clinicopathological significance. Methods:SP immunohistochemical method was used to detect the expression of RECK gene and MMP-9 protein in 60 specimens of oral squamous cell carcinoma and 10 normal oral epithelium tissues. And their relations with clinical pathology were analyzed. Results:The expression rates of RECK and MMP-9 protein were 51.67%and 90%, 78.33% and 20% in oral squamous cell cancer specimens and normal tissues respectively,the differences were significant (P〈0.05),and they were negatively correlated (r=-3.266,P〈0.05).The expression of RECK and MMP-9 protein in oral squamous cell carcinoma wasn’t correlated with the patient&#39;s age, gender and the tumor size etc. (P〉0.05). However, they were closely related to pathological grading, TNM stage and lymph node metastasis (P〈0.05) . Conclusion: The abnormal expression of RECK and MMP-9 may play important roles in the carcinogenesis and development of oral squamous cell carcinoma. And RECK gene may suppress the invasion and metastasis of oral squamous cell carcinoma through cutting down the MMP-9 protein expression.
