摘要
目的:观察转化生长因子-β1(transforming growth factor-β1,TGF-β1)对绒毛膜癌JEG-3细胞系基质金属蛋白酶-9(matrixmetalloproteinases,MMP-9)、基质金属蛋白酶抑制剂-1(TIMP-1)蛋白表达的影响。方法:体外培养人绒毛膜癌JEG-3细胞,分别给予不同浓度及不同作用时间的TGF-β1进行诱导,Western Blotting法检测MMP-9、TIMP-1蛋白的表达变化。结果:与对照组相比,100、200μg/L TGF-β1作用后,检测各组细胞MMP-9及TIMP-1蛋白的相对表达量均增加(P<0.01);与0h比较,12、24、48、72h各组细胞MMP-9及TIMP-1蛋白的相对表达量均增加(P<0.05),且各组MMP-9/TIMP-1大于1。结论:绒癌的浸润转移与MMP-9及TIMP-1的表达变化密切相关。
Objective: To investigate the effect of transforming growth factor-β1( TGF-β1) on matrixmetalloproteinases( MMP-9) and TIMP-1 protein expression in JEG-3 cells. Method: The human choriocarcinoma cell line cultured in vitro different doses of TGF-β1were given to JEG-3 cell at different time.The protein expression of MMP-9 and TIMP-1 induced by TGF-β1was measured by Western Blotting. Result: The protein expression of MMP-9 and TIMP-1 increased remarkedly,after inducing by TGF-β1with100μg / L and 200μg / L respectively,( P&lt;0.01). Conclusion: The invasive ability and metastasis of choriocarcinoma JEG-3 cells is closely related to the expression of MMP-9 and TIMP-1.
