摘要
目的探讨STAT-3与PTEN基因蛋白在子宫内膜癌及癌旁正常子宫内膜组织中的表达及意义,以及其在子宫内膜癌发生、发展中的作用。方法运用组织芯片技术以及免疫组织化学方法检测45例子宫内膜癌组织、30例癌旁正常子宫内膜组织中STAT-3和PTEN基因蛋白表达情况,并分析其与临床各参数间的关系,以及二者间的相关性。结果 STAT-3、PTEN在子宫内膜癌与癌旁正常子宫内膜组织间的阳性表达率有显著性差异(P均<0.05)。二者的阳性表达率与子宫内膜癌患者的肿瘤分化程度有关,与肿瘤临床分期、患者年龄、肌层浸润及绝经情况无显著相关性。子宫内膜癌组织中STAT-3与PTEN呈负相关。结论子宫内膜癌中STAT-3呈高表达,PTEN呈低表达,提示二者与子宫内膜癌的肿瘤分化程度相关,且两者之间有明显的相关性,表明子宫内膜癌的发生发展可能与STAT-3信号传导通路激活、细胞凋亡受抑制有关,而PTEN有抑制子宫内膜组织癌变的作用。
Objective It is to investigate the expression of STAT - 3 and PTEN protein in endometrial cancer of uterine and its clinicopathologic significance in order to detect if these proteins have effects on the development of endometrial carcinoma. Methods Tissue chip technique and immunohistochemical staining method were used to examine the expression of STAT3 and PTEN in 45 cases of endometrial carcinoma and in 30 cases of the normal endometrium. The relationship between these two subjects and their correlation with every clinical indexes was analyzed. Results There were significant differences in the positive rates of STAT3 and PTEN between endometrial carcinoma and normal endometrial tissue (P 〈0.05 ). The expression of STAT3 and PTEN had relation with histological grade in endometrial carcinoma, and had no significant correlation with clinical stage of the tumor, patient's age, muscle-invasion and menopause. Negative correlation was found between the expression of STAT3 and PTEN in endometrial carcinoma. Conclusion The over-expressions of STAT3 and the down-regulated expression of PTEN in endometrial carcinoma indicates that these two proteins are related with the histological grade, and they are closely correlated with each other, which shows that the progression of endometrial carcinoma may related with the activation of signaling pathways of STAT -3 and inhibition of apoptosis, and PTEN can inhibit the tumor's progression.
