摘要
目的探讨胃萎消颗粒对慢性萎缩性胃炎(CAG)大鼠胃黏膜的影响及其机制。方法选用SD大鼠,随机分为正常对照组、模型对照组和胃萎消颗粒低、中、高剂量组(剂量分别为1.25,2.5,5 g/kg)。除正常对照组外,其余各组均采用55℃的15%氯化钠溶液灌胃方法制作CAG动物模型。于造模后第13周开始给药治疗,至第24周末结束。观察胃黏膜组织萎缩、肠上皮化生、异型增生及损伤情况。采用免疫组化法检测大鼠胃黏膜细胞p53和增殖细胞核抗原(PCNA)蛋白的表达情况,用放射免疫法测定胃黏膜组织肿瘤坏死因子-α(TNF-α)的含量。结果 CAG模型大鼠胃黏膜组织p53阳性表达率、PCNA标记指数和TNF-α含量显著升高,经胃萎消颗粒治疗后,大鼠胃黏膜损伤指数明显降低,黏膜萎缩、肠上皮化生、异型增生鼠数明显减少,胃黏膜组织p53阳性表达率、PCNA标记指数和TNF-α含量明显降低(P<0.05或0.01)。结论胃萎消颗粒对CAG大鼠胃黏膜有保护作用,其机制可能与其抑制突变型p53基因、PCNA蛋白的表达和降低TNF-α等细胞因子活性有关。
Objective It is to discuss the effect of Weiweixiao granules on gastric mucosa of rats with chronic atrophic gastritis and its action mechanism. Methods SD rats were selected and randomly divided into normal control group, model control group and Weiweixiao granules with low, middle and high dose of 1.25, 2.5, 5 g/kg respectively. All groups except the normal control group were given 55℃, 15% sodium chloride solution by gavage method to establish CAG animal model. Weiweixiao granules treatment began on thirteenth week after making CAG animal model and ended at the end of the twenty-fourth week after making CAC animal model. Expression of p53 and PCNA protein of rat gastric mucosal cells in gastric mucosa was detected by immunohistochemistry, and content of TNF-α in gastric mucosa was determined by radioimmunoassay. Results p53 positive expression rates, PCNA labeling index and TNF-α contents increased significantly in gastric mucosa tissue of the CAG model, and the indexes decreased significantly (P 〈 0.05 or P 〈 0.01) with less mueosal atrophy, intestinal metaplasia,dysplasia after Weiweixiao granules treatment. Conclusion Weiweixiao granules has a protective effect on the gastric musosa of rats with CAG, and the mechanism may he related to the inhibition of the expression of mutant p53 gene, PCNA protein and decrease of TNF-α activity.
