贵州栽培灰毡毛忍冬的遗传多样性ISSR与SCoT标记 被引量:3

摘要 为了考察贵州栽培灰毡毛忍冬种源的遗传多样性水平,采用ISSR标记与SCoT标记分析方法对贵州省黔西县、绥阳县、金沙县和道真县的灰毡毛忍冬栽培类群进行遗传多样性分析。结果显示:1)ISSR标记与SCoT标记分析的多样性指数值趋势一致,在物种水平上的多态位点百分率(PPL)分别为72.73%和67.78%,基因分化系数(Gst)为0.329 7和0.415 0,栽培类群内的个体遗传差异大于类群间。2)在栽培类群间,黔西类群的遗传多样性水平、有效等位基因数高于其他类群,其次是绥阳类群,道真类群最低。3)遗传聚类显示,道真栽培类群独立为一类,黔西、绥阳与金沙类群聚为一类。SCoT标记方法更适合于灰毡毛忍冬种质资源遗传多样性的分子评价。 The genetic diversity of L.macranthoides cultivars among four groups of Qianxi,Suiyang, Jinsha and Daozhen in Guizhou was analyzed by ISSR and SCoT markers to explore the genetic diversity level of L.macranthoides cultivars in Guizhou.The results showed that the diversity index trend of L.macranthoides was almost consistent by using ISSR and SCoT markers.The percentage of polymorphic loci was 72.73% and 67.78% respectively at the species level,and the corresponding gene differentiation coefficient (Gst)was 0.329 7 and 0.415 0,which suggested that the genetic difference of intra-group was higher than inter-group.The level of genetic diversity (H)of Qianxi was 0.181 3,which was higher than other groups,the lowest group of H was DaoZhen(0.098 6).Cluster analysis based on genetic similarity indicated that the four groups ofL.Macranthoides cultivars could be classified into two classes.The L.Macranthoides of Daozhen group was the first class;the other class included Qianxi,Suiyang and Jinsha groups. SCoT marker was more suitable for molecular genetic diversity evaluation of L.macranthoides germplasm resources.
