摘要
目的建立测定复方三黄酊中2组分含量的高效液相色谱法。方法色谱柱:Thermo,ODS-2HYPERSIL,Dim(mm)250×4.6。流动相:甲醇-乙腈-0.025 mol/L磷酸溶液(用三乙胺调节pH为3.0)(15:15:70);流速:1.0ml/min;检测器:DAD检测器;检测波长:280nm和345nm;柱温:25℃。结果黄芩苷在0.06862 ug^0.6862 ug之间进样量与峰面积积分值呈良好的线性关系,平均加样回收率为99.42%,RSD=0.4%(n=6);盐酸小檗碱在0.0198~0.198 ug之间进样量与峰面积积分值呈良好的线性关系,平均加样回收率100.6%,RSD=1.9%(n=6)。结论本法简单易行,可用于复方三黄酊的含量测定。
Objective To establish the method for determination of the Baicalin and berberine Hydrochloride in Compound San -huang Tincture by HPLC.Methods HPLC chromat ographic condition:Thermo,ODS-2HYPERSIL,Dim( mm)250 ×4.6.The mobile phase consisted of methanol-acetonitrile -0.025 mol /L phosphoric acid solution ( adjusted with triethylamine to pH 3.0) (15:15:70) at a flow rate of 1.0 ml· min-1 .The DAD detection wavelength was 280nm and 235nm.The column temperature was 25℃.Re-sults The Baicalin was in good linearity over the range of 0.06862~0.6862 ug.The average recovery was 99.42%, with RSD of 0.4%(n=6).The berberine hydrochloride was in good linearity over the range of 0.0198 ~0.198 ug.The average recovery was 100.6%, with RSD of 1.9%(n=6).Conclusion This method is accurate,simple and can be used for the determination of Com-pound Sanhuang Chloramphenicol .
