摘要
为了探讨H19基因CpG岛甲基化变化对克隆羊及后代羊生长的影响,试验采用亚硫酸盐测序法分析成活克隆羊原代、后代与普通羊血细胞中H19基因CpG岛甲基化水平。结果表明:克隆羊原代(71.00%、70.00%)、后代(67.50%、68.50%)H19基因CpG岛甲基化水平与对照组(77.00%、66.00%)相比差异不显著(P>0.05),但对照组H19基因CpG岛甲基化水平显著差异(P<0.05),说明克隆羊与后代羊血液H19基因CpG岛甲基化水平接近,不会影响克隆动物及其后代的生长,但血细胞H19基因CpG岛甲基化水平与品种有关。
To investigate the changes of CpG island methylation of the H19 gene on the growth of the cloned sheep and their offspring, bisulfite sequencing was used to analyze detected the CpG island methylation levels of the H19 gene in the blood cells of the primary coned sheep and their offspring with ordinary sheep (control). The results indicated that there were not significant differences in the CpG islang methylation lev- els of the H19 gene of the primary coned sheep (71.00% ,70.00% ) and their offspring (67.50% ,68.50% ) compared with the control (77.00% ,66.00% ), but there was a significant difference in the CpG islang methylation level of the H19 gene of the control. The result indicate that the CpG islang methylation level of the H19 gene of the clone sheep is similar with that of their offspring, it will not affect the growth of them ,but the CpG islang methylation level of the H19 gene in the blood cells is associated with the breed.
