应用LAMP同时快速检测沙门菌和志贺菌的效果评价 被引量:4

Evaluation on simultaneous and rapid detection of Salmonella and Shigella by using LAMP

摘要 目的探讨环介导等温扩增(LAMP)方法同时快速检测沙门菌和志贺菌的应用价值。方法通过通用增菌培养,应用LAMP方法和分离培养法分别对76份食堂从业人员粪便标本进行沙门菌和志贺菌检测。结果LAMP方法检出1份标本沙门菌阳性,阳性率为1.32%;分离培养法检出1份都柏林沙门菌,阳性率为1.32%;2种方法的检测结果一致。LAMP方法可在3h内检出沙门菌、志贺菌,而分离培养法需要3-5d,鼠伤寒沙门菌LAMP方法的灵敏度为170CFU/ml,福氏志贺菌LAMP方法的灵敏度为190CFU/ml,与常见的食源性致病菌均无交叉反应。结论与分离培养法相比,LAMP方法特异性高,灵敏度高,不需要特殊仪器,能够在简易的实验条件下,快速、简便地同时检测沙门菌和志贺菌,LAMP方法适用于基层疾病控制实验室开展沙门菌和志贺菌的快速筛查检测。 [ Objective] To discuss the application value of loop-mediated isothermal amplification (LAMP) in simultaneous and rapid detection of Salmonella and Shigella. [ Methods ] Through the common enrichment culture, LAMP method and isolation meth- od were used for detection of Salmonella and Shigella in stool specimens of 76 copies of the canteen staff. [ Results ] The LAMP method detected 1 samples positive for Salmonella, the positive rate was 1.32%. Isolation and culture method was used to detect 1 Salmonella Dublin, the positive rate was 1.32%. The results of 2 methods were consistent. LAMP method can detect Salmonella and Shigella within 3 hours, while isolation and culture method needed 3-5 days. The sensitivity of LAMP for Salmonella and Shi- gella detection was 170 CFU/ml and 190 CFU/ml,respectively. No cross reactivity was observed between common foodborne patho- gens and Salmonella, Shigella. [ Conclusion] Compared with the isolation and culture method, LAMP method has high specificity, high sensitivity, no special instrument. Under the simple experimental conditions, LAMP method can detect Salmonella and Shigella simultaneously and rapidly, it is suitable for rapid screening and detection of Salmonella and Shigella by grassroots CDC laboratory.
