摘要
目的:为了观察姜黄素对人肥大细胞系HMC-1细胞胸腺基质淋巴细胞生成素(TSLP)表达水平的抑制作用。方法:采用ELISA、定量RT-PCR、免疫印迹及caspase-1活性检测等实验来反映姜黄素的作用。结果:姜黄素能抑制HMC-1细胞TSLP的产生及mRNA的表达,50μmol/L姜黄素的最大抑制率能达到29.5%±5.3%。姜黄素对PMA与A23187诱导的核内NF-κB p65表达也有抑制。此外,降低在激活HMC-1细胞中显著增加的caspase-1活性。结论:本研究揭示了姜黄素可抑制TSLP的表达,对炎症及特应性疾病等有潜在的治疗作用。
Objective:To investigate the inhibitory effect of curcumin on the expression and production of TSLP in the human mast cell line, HMC-1 cells. Methods: Using enzyme-linked immnnosorbent assay, reverse transcription-polymerase chain reaction, Western blot, and caspase-1 assay to investigate the effects of curcumin. Results: Curcumin inhibited the production and mRNA ex- pression of TSLP in HMC-1 cells, the maximal inhibition rate of TSLP production by 50 μmol/L curcumin was 29.5% ± 5.3%. Also, curcumin suppressed the nucleus expression of nuclear factor-κB (NF-κB) p65 induced by PMA plus A23187. In addition, easpase-1 activity was decreased obviously by pretreatment with curcumin in the activated HMC-1 cells. Conclusion: These results suggest that curcumin can be used to treat inflammatory and atopic diseases through the suppression of TSLP.
