两种线栓制备法制作大鼠大脑中动脉阻塞再灌注损伤模型的研究 被引量:2

摘要 目的评价两种制备线栓的方法,为脑缺血实验研究提供稳定可靠的动物模型。方法 30只SD雄性大鼠(250~280 g)随机分成两组各15只,第一组采用Zea Longa流派(线栓头端加热成球形),第二组采用Koizumi流派(线栓头端用胶包裹),两种线栓制备法分别制作大鼠大脑中动脉阻塞再灌注损伤模型,观察手术时间、神经功能评分,计算梗死百分比,并分析神经功能评分和脑梗死体积比之间的关系,对两种线栓制备法模型的成功率与稳定性进行评价。结果两组在神经功能评分、脑梗死体积比差异无统计学意义(P>0.05);两组模型的稳定性相近;神经功能评分与脑梗死体积比之间存在相同变化趋势;第二组操作时间少于第一组,两组之间的差异具有统计学意义(P<0.01)。结论第二组的线栓制备法线栓头采用密封胶包裹,线栓头大小容易控制,较第一组线栓头端加热成球形所需时间短,且易操作。 Objective To evaluate the effect of two methods of making suture-embolus, and to provide more stable and reliable animal models for experimental study of cerebral ischemia. Methods Thirty male Sprague-Dawley (SD) rats (250-280 g) were randomly divided into two groups, with 15 rats in each group. The rats of group one were subjected to temporary MCAO/R by using the method ofZea Longa (the tip of suture-embolus be heat- ed to round), and those of group two were subjected to Koizumi (the tip of suture-embolus be wrapped by sealant). By recording the time of making MCAO/R model, the grade of neurobehavioral function of rats, calculating the infarct vol- ume ratios and analyzing the relationship between the latter two, the success ratio and stability of two methods of mak- ing suture-embolus were evaluated. Results There was no significant difference on the evaluation of neurobehavioral function and on the volume ratios of brain infarction. The stability of the two models was similar. There was the same trend between the neurobehavioral function and the volume of brain infarction. The operate time of group two is shorter than group one. The difference between two groups has statistical significance (P〈0.01). Conclusion The method for group two (the tip of suture-embolus can be wrapped by sealant, and its size can be controlled more easily) is easier to operate and need shorter time than the method for group one (the tip of suture-embolus be heated to round).
