摘要
目的建立测定肺癌小鼠血浆及各脏器组织中紫杉醇含量的HPLC法。方法血浆及组织样品经萃取后,以甲醇-乙腈-醋酸铵溶液(47.5∶32.5∶20)为流动相,使用phenomenex C18色谱柱(250mm×4.6mm,5μm)进行分离,柱温为30℃,UV检测波长为230nm,内标物为多烯紫杉醇。结果血浆中紫杉醇的线性范围为0.02606-2.2606μg/mL,肺中紫杉醇的线性范围为1.015-203.00μg/g,脾肝肾中紫杉醇的线性范围为0.203-20.3μg/g,心脏中紫杉醇的线性范围为0.203-10.15μg/g。提取回收率>90%,日内、日间变异系数<10%。结论内标标准曲线法灵敏度高、重复性好、操作简便、快速,可作为生物样品中紫杉醇浓度的测定及药代动力学研究的分析方法。
Objective To develop an HPLC method for quantitative determination of paelitaxel in rat blood plasma and tissue. Methods Blood Plasma and tissue samples were extracted with methanol. The residues discovered were analyzed with a reverse phase HPLC system C18 column, 4.6mm × 150mm, 5μm). Mobile phase was consisted of methanol- acetonitrile - ammonium acetate (47.5: 32.5: 20) and UV detection was set 230nm. Results The linear range of paclitaxel in plasma was 0. 02606 - 2. 2606μg/mL, the lung 1. 015 - 203. O0 Ixg/g, the spleen kidney 0.203 -20.30 μg/g, the heart O. 203 - 10.15 μg/g. The recoveries for paclitaxel was more than 90%. The inter -day and intra- day relative standard deviations was smaller than 5%. Conclusions The method provides a sensitive, accurate, precise and reliable analytical procedure for pharmacokinetic studies of paclitaxel in rat.
