摘要
为了探讨猪卵巢采集月份、保存温度及保存时间对卵母细胞体外成熟的影响,对不同月份、不同保存温度及不同保存时间的猪卵巢采用抽吸法获得卵丘卵母细胞复合体(COCs),体外培养统计其第一极体排出情况.结果表明,从屠宰场所取卵巢的COCs数5~7月和8~10月期间显著(P<0.05)高于2~4月和11月至次年1月,而且其卵母细胞体外核成熟率在5~7月和8~10月期间也显著(P<0.05)高于2~4月和11月至次年1月;采集的猪卵巢分别置于15、20、25、30和35℃的生理盐水中2h,收集卵巢的COCs数差异不显著(P>0.05),但是存放在30和35℃的生理盐水中的卵巢的卵母细胞体外成熟率极显著(P<0.01)高于其他3组;置于35℃生理盐水中2、4、6、8h的卵巢的COCs数差异不显著(P>0.05),但存放2h的卵巢的卵母细胞体外成熟率显著(P<0.05)高于存放4、6和8h的.
The effects of ovarian collecting month,storage temperature and time on mature of pig oocytes in vitro were investigated through the statistics of culture of oocytes in vitro obtained by aspiration method and the extruding of the first polar body.The results showed that the number of the cumulus-oocyte complexes (COCs) and the maturation rate of oocytes in vitro during May to July and August to October were significantly higher than those during February to April and November to January.The number of the cumulus-oocyte complexes (COCs) had no significant difference if the collected ovary was stored in physiological saline at 15 ℃,20 ℃,25 ℃,30℃ and 35℃ for 2h (P> 0.05).But the maturation rate of oocytes in vitro from the ovary stored at 30℃ and 35 ℃ were significantly higher than that of the other 3 groups (P<0.01).The number of the cumulus-oocyte complexes(COCs) had no significant difference when the ovary was placed at 35 ℃ in physiological saline for 2 h,4 h,6 h and 8 h (P> 0.05),but the maturation rate of oocytes in vitro from ovaries stored for 2 h were significantly higher than that of the other storage groups(P< 0.05).
