摘要
以不同来源的槲蕨[Drynaria fortunei(Kunze)J.Sm]孢子、块茎、叶片为试验材料,在培养基中加入不同浓度的6-BA、2,4-D、NAA、IBA等激素,研究植物生长调节剂对槲蕨离体培养的效应。结果表明,适当浓度的2,4-D对槲蕨愈伤组织诱导有良好的促进作用,且愈伤组织较大、嫩绿、质量好,愈伤组织诱导最佳激素组合为1.0 mg/L 6-BA+1.0 mg/L 2,4-D,诱导率为53.3%;最佳分化培养基为1.0 mg/L 6-BA+0.5 mg/L KT,分化率为75.0%;继代增殖中6-BA与NAA配比有较好的效果,最适增殖培养基为2.0mg/L 6-BA+0.5 mg/L NAA,最高增殖倍数可达2.25倍;最佳生根壮苗培养基为1/2 MS+0.5 mg/L NAA+0.3 mg/L IAA。
Spores,rhizomes and leaves of Drynaria fortunei (Kunze) J.Sm were used to investigate effects of medium with different concentration hormone of 6-BA,2,4-D,NAA,IBA on culture in vitro.The results showed that suitable concentration of 2,4-D could facilitate the callus induction of Drynaria fortunei (Kunze) J.Sm.The callus is big and green with good quality.The best hormone combination for callus induction was 1.0 mg/L 6-BA + 1.0 mg/L 2,4-D with the highest callus induction rate of 53.3%.The best differentiation medium was 1.0 mg/L 6-BA + 0.5 mg/L KT with the highest differentiation rate of 75.0%.In the culture of multiplication,the best combination was 6-BA and NAA,and the best recipe of multiplication culture medium was 2.0 mg/L 6-BA +0.5 mg/L NAA with the highest proliferation times of 2.25.The best rooting medium was 1/2 MS+0.5 mg/L NAA+0.3 mg/L IAA.
